Direct CHO Host cell protein contaminant quantification  in bioprocess operations

CHO Host Cell Proteins (HCP) are contaminants originating from the CHO cells used to manufacture therapeutic antibodies. The HTRF® CHO HCP kit can be used for the quantification of CHO HCP proteins in routine bioprocess operations, from the very crude harvest material to the final product. The simple and robust procedure benefits from increased throughput compared to ELISA.

Assay principle

The HTRF® CHO Host Cell Protein (HCP) assay is a sandwich immunoassay for the measurement of CHO HCP. The assay uses two specific polyclonal antibodies, one labelled with Eu3+-Cryptate and the other with d2. The specific signal is proportional to the concentration of CHO HCP present in the sample. This homogeneous assay requires only a single reagent addition (by premixing detecting reagents) and 2 hours to ON incubation for the quantitative detection of CHO HCP.

Product specifications

HTRF® CHO HCP assay can be run on whole cells or in cell supernatant using the protocol described below. The antibodies labelled with HTRF® fluorophores may be pre-mixed and added in a single dispensing step to further streamline the protocol.  The assay can be run in 96-  to 384-well plates by simply resizing each addition volume proportionally. The signal can be read after 2 hours incubation at RT. Maximum sensitivity is obtained with an ON incubation.

Detection Limit: 2 ng/mL
Dynamic Range: 2 to 100 ng/mL
Incubation Time: ON at RT (2H with lower sensitivity)

HTRF technology allows multiple readout using the same plate. Figure 1 describes CHO HCP calibration curve in 20µL final using a white 384 well small volume plate. Readout on PHERAstar FS (Laser) was performed after 2H and ON incubation period.
