HTRF® assay kit for detecting GAD67 in cell lysates

The GAD67 HTRF® kit is a rapid sandwich immunoassay optimized for the quantification of the glutamate decarboxylase 67kDa isoform (product of the GAD1 gene) in cell lysates. Developed with the add-and-read no-wash HTRF technology, the kit represents a significant improvement over the traditional ELISA protocol, resulting in major time savings. The kit lysis buffer has been optimized for full protein extraction. The kit combines high detection performance with low sample consumption, making it the ideal solution for laboratories looking for a reliable and streamlined detection of human, rat or mouse GAD67.

 

Assay principle

GAD67 is measured using a sandwich immunoassay of two antibodies specific for GAD67, respectively labelled with Terbium cryptate (donor fluorophore) and d2 (acceptor fluorophore). The intensity of the FRET signal is proportional to the concentration of the GAD67 present in the sample.

The GAD67 assay protocol is shown here. Cells are plated, stimulated and next lysed in the same 96-well culture plate. Lysates are then transferred to the assay plate for the detection of GAD67 by HTRF® reagents. This protocol enables the cells' viability and confluence to be monitored. The antibodies labelled with HTRF fluorophores may be pre-mixed and added in a single dispensing step, to further streamline the assay procedure. The assay detection can be run in 96- to 384-well plates by simply resizing each addition volume proportionally.
