HTRF® assay kit for Friedreich's ataxia research

The HTRF® human frataxin assay kit enables the rapid and accurate quantification of frataxin in cell lysates. The protocol is based on a sandwich immunoassay, using two specific antibodies. The streamlined add & read protocol is suitable for miniaturization, requiring only small sample volumes. The simple and robust procedure of this kit is amenable for any use from basic research to all drug screening phases, providing increased throughput compared to technologies such as ELISA. The HTRF® frataxin assay is designed for CNS investigation, particularly for Friedreich's ataxia research.

Assay principle

Human frataxin is measured using a sandwich immunoassay involving two specific frataxin antibodies, respectively labelled with Europium cryptate (donor) and d2 (acceptor). The intensity of the signal is proportional to the concentration of frataxin present in the sample.

The human frataxin assay protocol is described here. Cells are plated, stimulated and next lysed in the same 96-well culture plate. Lysates are then transferred to the assay plate for the detection of human frataxin by HTRF® reagents. This protocol enables the cells' viability and confluence to be monitored. The antibodies labelled with HTRF fluorophores may be pre-mixed and added in a single dispensing step, to further streamline the assay procedure. The assay detection can be run in 96- to 384-well plates by simply resizing each addition volume proportionally.
