Assays for measuring phosphorylated and total proteins directly in cell

HTRF® cellular phospho-/total protein assays provide precise, sensitive measurement of endogeneous levels of phosphorylated and non phophorylated proteins able to be achieved in an endogenous cellular environment. After lysis of the cell membrane, phospho- and total proteins can be detected using the kit reagents. All assays are based on the same sandwich immunoassay construction, comprising two specific monoclonal antibodies coupled to HTRF donor and acceptor dyes. Due to their high sensitivity, the assays are suitable for either over-expressed or endogenous receptors, and can be used in many types of cells including primary cells. Being truly homogeneous, they represent a robust, reliable and rapid alternative to more conventional technologies such as Western Blot, ELISA or other luminescent bead-based assays, and can be used from basic research up to High Throughput drug screening for both small molecule and biologic investigation.
