Immune checkpoint assay to identify LAG3/MHC II checkpoint inhibitors

The lymphocyte activation gene-3 (LAG-3), selectively transcribed in human activated T and NK cells, encodes a ligand for major histocompatibility complex (MHC) class II molecules. When LAG3 is bound to MHC II, T cell response is suppressed. Checkpoint inhibitors blocking LAG3/MHC II complex formation are generating considerable interest in cancer immunotherapy.

The HTRF LAG3/MHC II Binding Assay is designed to measure the interaction between Human LAG3 and MHC II proteins. Utilizing HTRF (Homogeneous Time-resolved Fluorescence) technology, the assay enables simple and rapid characterization of compound and anti-human antibody inhibitors in a high throughput format.

Our kits include everything you need to set up your biochemical test: validated recombinant proteins, labelled detection antibodies, and buffers. They can be used in all formats from 96 to 1536 wells, and have been validated with reference antibodies and compounds.

Assay principle

HTRF® - The homogeneous biochemical immunoassay

Each kit includes recombinant immune checkpoint partners tagged for detection.

Without treatment, the partners interact and the binding of each detection antibody to its tag target generates an HTRF signal.

Inhibitory effects of blocking mouse and human mAbs

The inhibitory effects of human and mouse anti-LAG3 were tested at 10 nM LAG3 and 20 nM MHC II. Assay windows around 3 were obtained.
