HTRF® assay kit for detecting human mesencephalic astrocyte-derived neurotrophic factor in cell supernatants

Based on our TR-FRET homogeneous and robust HTRF® technology, the human MANF sandwich immunoassay kit is designed for the rapid detection of endogeneous mesencephalic astrocyte-derived neurotrophic factor in cell supernatants. Using a streamlined no-wash protocol, this simple and robust assay is suitable for miniaturization and enables high throughput operations. The low sample sizes (down to 5 µL) mean precious biological samples can be saved.

Assay principle

Human MANF is measured using sandwich immunoassays involving two specific human MANF antibodies, respectively labelled with Europium cryptate (donor) and d2 (acceptor). The intensity of the signal is proportional to the concentration of MANF present in the sample.

The human MANF assay protocol is described here, using a 384-well small volume white plate (20 µL final). Cell supernatant, sample or standard are dispensed directly into the assay plate for the detection of human MANF by HTRF® reagents. The antibodies labelled with HTRF fluorophores may be pre-mixed and added in a single dispensing step, to further streamline the assay procedure. The assay can be run in 96- to 384-well plates by simply resizing each addition volume proportionally.
