Sandwich immunoassay kits designed for the rapid detection of human or mouse Utrophin

Based on our TR-FRET homogeneous robust HTRF® technology, the human utrophin and mouse utrophin sandwich immunoassay kits are designed for the rapid detection of utrophin in cell lysates. This simple and robust procedure is suitable for miniaturization and enables work with low sample sizes. The add & read protocol eliminates all wash steps for faster analysis. By detecting utrophin, also known as dystrophin-related protein 1, this HTRF® kit is a critical tool for research into Duchenne's muscular dystrophy.

Assay principle

Human utrophin or mouse utrophin are measured using sandwich immunoassays involving two specific anti-utrophin antibodies, respectively labelled with Europium cryptate (donor) and d2 (acceptor). The intensity of the signal is proportional to the concentration of utrophin present in the sample.

The human or mouse utrophin assay protocol are described here. Cells are plated, stimulated and next lysed in the same 96-well culture plate. Lysates are then transferred to the assay plate for the detection of utrophin by HTRF® reagents. This protocol enables the cells' viability and confluence to be monitored. The antibodies labelled with HTRF fluorophores may be pre-mixed and added in a single dispensing step, to further streamline the assay procedure. The assay detection can be run in 96- to 384-well plates by simply resizing each addition volume proportionally.
