HTRF® assay kit for detecting human PCSK-9 in cell supernatants

Based on our TR-FRET homogeneous and robust HTRF® technology, the human PCSK-9 sandwich immunoassay kit is designed for the detection of proprotein convertase subtilisin/kexin type 9 directly in cell supernatants. The simple and rapid protocol adapts to automation and miniaturization, and enables high throughput operations. Using this assay, human PCSK-9 can be directly detected in your cell type of interest, such as liver cells.

Assay principle

Human PCSK-9 is measured using sandwich immunoassays involving two specific human PCSK-9 antibodies, respectively labelled with Europium cryptate (donor) and d2 (acceptor). The intensity of the signal is proportional to the concentration of the PCSK-9 present in the sample.

The PCSK-9 assay protocol is described here, using a 384-well small volume white plate (20 µL final). Cell supernatant, sample or standard are dispensed directly into the assay plate for the detection of PCSK-9 by HTRF® reagents. The antibodies labelled with HTRF fluorophores may be pre-mixed and added in a single dispensing step, to further streamline the assay procedure. The assay can be run in 96- to 384-well plates by simply resizing each addition volume proportionally.
