Cell-based assay for phospho-TAU measurement

Tau exists in different states in both Alzheimer’s (AD) and Parkinson’s (PD) diseases. Tau hyperphosphorylation is also a marker for multiple neurodegenerative diseases and CNS disorders.

As all Tau modifications are relevant in biological terms, Cisbio developed a complete range of kits to study the Tau family.

The HTRF Tau phosphorylation (Ser202/Thr205) assay is designed for rapidly detecting human phosphorylated Tau in brain tissues and cell cultures. It can be used in combination with our Map Tau assay, which measures total Tau protein for normalization against steady state expression.

Assay principle

HTRF® - the homogeneous cell-based sandwich immunoassay

All of our Tau immunoassays were developed in HTRF sandwich format with specific antibodies bound to either donor (cryptate) or acceptor (d2) fluorophores, which enables energy transfer when they bind to the same target protein.

In this assay, antibodies are directed against specific phospsorylation sites on Tau protein, and the specific signal generated is proportional to the level of phosphorylation.

This protocol requires just a single addition by pre-mixing detection reagents. It can be run in one plate or with a transfer step where cells are cultivated and treated in the preferred format (6 wells, 96 wells …), then lysed and transferred to a 384-well plate for the detection step.

Product Performance

Detection of phosphorylated Tau in transgenic mouse brain extracts

Triple-transgenic mice (Tau / PSEN2 / APP) were used as Alzheimer models. These mice develop an Alzheimer-like pathology that starts at 10 months old and progresses steadily over time, according to behavioral studies.

The earlier stages of AD development in this model are difficult to monitor in a cell-based assay, since Tau phosphorylation is at very low levels.

The aim of the experiment was to detect previously unseen differences between 10 and 14 month-old mice.

As the graph shows, Cisbio’s Tau phosphorylation kit enables clear discrimination of all stages.
